ABSTRACT

Objective: To study the incidence of bloodstream infections, pathogen distribution, and antibiotic resistance profile in patients with hematological malignancies. Methods: From January 2018 to December 2021, we retrospectively analyzed the clinical characteristics, pathogen distribution, and antibiotic resistance profiles of patients with malignant hematological diseases and bloodstream infections in the Department of Hematology, Nanfang Hospital, Southern Medical University. Results: A total of 582 incidences of bloodstream infections occurred in 22,717 inpatients. From 2018 to 2021, the incidence rates of bloodstream infections were 2.79%, 2.99%, 2.79%, and 2.02%, respectively. Five hundred ninety-nine types of bacteria were recovered from blood cultures, with 487 (81.3%) gram-negative bacteria, such as Klebsiella pneumonia, Escherichia coli, and Pseudomonas aeruginosa. Eighty-one (13.5%) were gram-positive bacteria, primarily Staphylococcus aureus, Staphylococcus epidermidis, and Enterococcus faecium, whereas the remaining 31 (5.2%) were fungi. Enterobacteriaceae resistance to carbapenems, piperacillin/tazobactam, cefoperazone sodium/sulbactam, and tigecycline were 11.0%, 15.3%, 15.4%, and 3.3%, with a descending trend year on year. Non-fermenters tolerated piperacillin/tazobactam, cefoperazone sodium/sulbactam, and quinolones at 29.6%, 13.3%, and 21.7%, respectively. However, only two gram-positive bacteria isolates were shown to be resistant to glycopeptide antibiotics. Conclusions: Bloodstream pathogens in hematological malignancies were broadly dispersed, most of which were gram-negative bacteria. Antibiotic resistance rates vary greatly between species. Our research serves as a valuable resource for the selection of empirical antibiotics.

ABSTRACT

Character strengths could efectively prevent negative psychological outcomes in adults. However, there was little research conducted among early adolescents. The present study aimed to explore character strengths that were independently related to fewer behavior problems in early adolescents. In total, 521 early adolescents (mean age 10.92 ± 0.04, range 10­12 years) were recruited from primary schools in Sichuan, China. Character strengths were measured using the Values in Action Inventory of Strengths for Youth (VIA-Youth). Behavior problems were measured using the Conners Parent Symptom Questionnaire (PSQ). The results showed that behavior problems were negatively correlated with character strengths (r = −0.14 to −0.3, p < 0.05 Bonferroni corrected). Character strengths explained a signifcant proportion of additional variance (14­22%) in fve types of behavior problems after controlling the efect of demographic factors (residence, left-behind experiences, maternal education level). Moreover, several specifc character strengths showed an independent contribution (ß = −0.34 to −0.14 for self-regulation, perseverance, zest, humility, and leadership; ß = 0.21 to 0.34, for hope; all p < 0.05) to behavior problems. Our study revealed that character trengths were protective factors against behavior problems in early adolescents.

ABSTRACT

OBJECTIVE@#To evaluate of the clinical effects of mobile-bearing(MB) and fixed-bearing(FB) unicompartmental knee arthroplasty(UKA) in the treatment of knee osteoarthritis by Meta-analysis.@*METHODS@#The literature on FB UKA and MB UKA in the treatment of knee osteoarthritis in PubMed, CNKI, Wanfang, Cochrane and EMBASE database were searched by computer from January 2000 to April 2020. According to the inclusion and exclusion criteria, two authors were selected independently and the selected literature was evaluated for quality.After literature data were extracted, Review Manager 5.3 software was used to analyze knee function score, postoperative activity, revision rate, polyethylene wear rate, pad dislocation, aseptic loosening, postoperative pain, knee arthritis progression, mechanical shaft alignment of lower limbs, and imaging clarity line respectively.@*RESULTS@#A total of 13 literatures were included in this meta-analysis, including 2 randomized controlled studies and 11 cohort studies. A total of 1 871 patients were included, including 913 in FB UKA group and 958 in MB UKA group. Meta analysis results showed that:postoperative knee joint function score[MD=-0.84, 95%CI(-1.46, -0.21), P=0.008] and postoperative knee joint range of motion [MD=-1.51, 95%CI(-2.84, -0.18), P=0.03] in FB UKA group were better than those in MB UKA group. Compared with FB UKA group, MB UKA group had a higher lower limb mechanical axis alignment rate[OR=2.08, 95%CI(1.27, 3.39), P=0.003], and the wear rate of polyethylene [OR=0.11, 95%CI(0.01, 0.91), P=0.04] was lower. There were no differences between two groups in the renovation rate [OR=1.16, 95%CI(0.75, 1.80), P=0.50), liner dislocation rate[OR=3.78, 95%CI(0.93, 15.29), P=0.06], aseptic loosening [OR=2.11, 95%CI(0.81, 5.51), P=0.13], postoperative pain[OR=1.13, 95%CI(0.37, 3.43), P=0.83], osteoarthritis progression[OR=1.28, 95%CI(0.67, 2.47), P=0.46)and imaging radiolucent line[OR=1.62, 95%CI(0.09, 30.22), P=0.75].@*CONCLUSION@#FB UKA has a higher postoperative functional score and range of motion.MB UKA has more advantages in the correction of lower limb mechanical axis, and the wear rate of polyethylene is also lower. There was no significant difference between the two groups in revision rate, dislocation of the liner, aseptic loosening, postoperative pain, progression of osteoarthritis, and postoperative translucency.

ABSTRACT

Objective:To evaluate the role of Toll-like receptor 4 (TLR4) in the mechanism by penehyclidine hydrochloride alleviating lipopolysaccharide (LPS)-induced acute lung injury(ALI)in rats.Methods:Twenty-four healthy male Sprague-Dawley rats, aged 10 weeks, weighing 220-250 g, were divided into 4 groups ( n=6 each) according to the random number table method: normal saline group (NS group), endotoxin-induced ALI group (ALI group), penehyclidine hydrochloride + normal saline group (PHC+ NS group) and penehyclidine hydrochloride + endotoxin-induced ALI group (PHC+ ALI group). ALI was induced by intratracheal instillation of LPS 5 mg/kg in anesthetized animals. In group PHC+ ALI, penehyclidine hydrochloride 2 mg/kg was intraperitoneally injected immediately after intratracheal instillation of LPS. The equal volume of normal saline was injected into the airway in group NS, and penehyclidine hydrochloride 2 mg/kg was intraperitoneally injected immediately after intratracheal instillation of normal saline in group PHC+ NS. The rats were sacrificed at 6 h after intratracheal instillation of LPS or normal saline, and lungs were removed. The lung was lavaged and broncho-alveolar lavage fluid (BALF) was collected for determination of concentrations of tumor necrosis factor-alpha (TNF-α) and interleukin-1beta (IL-1β) by enzyme-linked immunosorbent assay. Lung tissues were obtained for microscopic examination of the pathological changes (with a light microscope) and for determination of wet to dry lung weight ratio (W/D ratio) and TLR4 protein and mRNA expression (by immuno-histochemistry or real-time polymerase chain reaction). Results:Compared with group NS and group PHC+ NS, the W/D ratio and concentrations of TNF-α and IL-1β in BALF were significantly increased, the expression of TLR4 protein and mRNA in lung tissues was up-regulated ( P<0.01), and the pathological changes of lung tissues were aggravated in group ALI . Compared with group ALI, the W/D ratio and concentrations of TNF-α and IL-1β in BALF were significantly decreased, the expression of TLR4 protein and mRNA in lung tissues was down-regulated ( P<0.01), and the pathological changes of lung tissues were significantly mitigated in group PHC+ ALI. Conclusions:The mechanism by which penehyclidine hydrochloride reduces endotoxin-induced lung injury may be related to reduction of TLR4 activity and thus inhibition of pulmonary inflammatory responses in rats.

ABSTRACT

Objective To analyze the infiltration abundance of macrophage M2 in breast cancer tissues and explore the correlation between VSIG4 and macrophage M2 and the potential mechanism of regulating the invasion and migration of breast cancer patients. Methods We downloaded the RNA-seq data of TCGA-BRCA and assessed the infiltration abundance of immune cells in the samples by CIBERSORT, and established a prognostic risk prediction model. Then, we analyzed the effect of macrophage M2 and VSIG4 on the prognosis of breast cancer patients. In addition, we analyzed the signaling pathway associated with VSIG4 by gene set enrichment analysis and predicted its upstream regulation of miRNA. Results The infiltration abundance of macrophage M2, age, PR status and pathological stage were involved in the establishment of risk prediction model, and the model had a good prediction performance (AUC=0.816). High infiltration of macrophage M2 (HR=1.35, P < 0.05) and high expression of VSIG4 (HR=1.4, P=0.039) suggested poor prognosis of breast cancer patients. VSIG4 could be regulated by upstream miR-29a-3p and significantly correlated with Toll-like receptor, cell adhesion, production and release of cytokine. Conclusion VSIG4 is significantly associated with breast cancer patients' prognosis and infiltration of macrophage M2, regulated by the upstream miR-29a-3p and promotes the invasion and migration of breast cancer cells. It can be used as a potential prognostic marker for breast cancer.

ABSTRACT

Objective:To investigate the effect of suspended leukocyte poor red blood cell transfusion on hemeoxygenase-1 (HO-1) expression and T lymphocyte subsets in peripheral blood of patients with myelodysplastic syndrome.Methods:50 cases of MDS patients treated in Shandong Blood Center from January 2016 to December 2019 were selected as the research object. According to the different infusion of blood components, they were randomly divided into the observation group and the control group, with 25 cases in each group.The patients in the observation group were treated with s suspended leukocyte poor red blood cell transfusion, and the patients in the control group were treated with washing red blood cell infusion.The level of HO-1, interleukin-6 (IL-6) and TNF-α and the T cell subset such as CD 3+, CD 4+, CD 8+ and CD 4+/CD 8+ in the serum before and after transfusion were tested by enzyme linked immunosorbent assay and flow cytometry, respectively. Results:There was no statistically significantdifference in the level of HO-1, IL-6 and TNF-α in the serum of the two groups before transfusion ( P>0.05); After transfusion, TNF-α in the serum of the observation group significantly decreased, compared with that in control group: (152.10 ± 21.89) ng/L vs. (201.14 ± 28.90) ng/L, (1.34 ± 0.45) ng/L vs. (2.89 ± 1.01) ng/L. However, the HO-1 significantly increased: (78.91 ± 15.74) μg/L vs. (58.99 ± 13.33) μg/L, andthe difference was statistically significant ( P<0.05). There was nostatistically significantdifference in the immunologic functions of the two groups before transfusion ( P>0.05); After transfusion, CD 4+/CD 8+of the observation group both significantly increased, compared with that inthe control group: (49.11 ± 19.13)% vs. (47.13 ± 12.84)%, (25.23 ± 10.80)% vs. (21.09 ± 12.28)%, (24.74 ± 10.84)% vs. (21.88 ± 11.18)%, 1.02 ± 0.25 vs. 0.96 ± 0.20, and the difference was statistically significant ( P<0.05). Conclusions:The expression level of HO-1 in peripheral blood and immune function of patients with myelodysplastic syndrome can be improved by suspended leukocyte poor red blood cell transfusion.

ABSTRACT

Isogenic cells growing in identical environments show cell-to-cell variations because of the stochasticity in gene expression. High levels of variation or noise can disrupt robust gene expression and result in tremendous consequences for cell behaviors. In this work, we showed evidence from single-cell RNA sequencing data analysis that microRNAs (miRNAs) can reduce gene expression noise at the mRNA level in mouse cells. We identified that the miRNA expression level, number of targets, target pool abundance, and miRNA-target interaction strength are the key features contributing to noise repression. miRNAs tend to work together in cooperative subnetworks to repress target noise synergistically in a cell type-specific manner. By building a physical model of post-transcriptional regulation and observing in synthetic gene circuits, we demonstrated that accelerated degradation with elevated transcriptional activation of the miRNA target provides resistance to extrinsic fluctuations. Together, through the integrated analysis of single-cell RNA and miRNA expression profiles, we demonstrated that miRNAs are important post-transcriptional regulators for reducing gene expression noise and conferring robustness to biological processes.

Objective@#To explore the effect of orienteering exercises on the improvement of children s executive function, and to explore the relationship between executive function and orienteering intervention to provide theoretical support.@*Methods@#Forty children from the fourth grade of Zhonghai the First Experimental Primary School in Changchun City were selected as the experimental subjects, 20 as the experimental group (10 males, 10 females) and 20 as the control group (10 males, 10 females). The functional changes of executive function subfunctions (inhibitory function, conversion function, refresh function) before and after orienteering exercises intervention were measured by More odd shifting, 1 back and Flanker.@*Results@#After intervention, the inhibitory function, conversion function and refresh function were significantly decreased in the experimental group (10.29±15.99, 295.19±189.76, 642.85±220.78)ms compared with before intervention (25.62±10.18, 616.04±287.92, 1 051.25±275.00)ms (F=12.52, 20.76, 20.89, P<0.01), while there was no significant change in the control group (P>0.05). In this study, neither gender main effect nor interaction between sex × group, sex × time and sex × time × group were found (P>0.05).@*Conclusion@#Orienteering exercises can significantly improve children s executive function, which are not vaired by sex.

ABSTRACT: Herbicide application is an effective weed control method for mitigating crop yield loss; however, herbicide overuse can cause toxicity in non-target plants. The present study evaluated the effects of glufosinate at recommended dose for agricultural application (0.45 kg ha-1) and at overuse dose (0.90 kg ha-1) glufosinate application on photosynthetic performance and nitrogen assimilation of the rapeseed varieties D148 and Zhongshuang 11 (ZS11). Both glufosinate concentrations significantly decreased the content of chlorophyll and nitrogenous compounds, except free proline, and the activity of glutamine synthetase and glutamate synthase, and increased the activity of glutamic acid dehydrogenase in both varieties. When the concentration of glyphosate was 0.45kg ha-1, the nitrogen assimilation of the two varieties decreased, which indicated that the recommended dosage inhibited the nitrogen assimilation of the two varieties; however, the increase of net photosynthetic rate of D148 and the decrease of that of ZS11 mean that D148 is more tolerant to the recommended dose of glyphosate than ZS11. The 0.90 kg ha-1 dosage was toxic to both rapeseed varieties. Overall, our results indicated that herbicide overuse inhibited the photosynthetic rate and nitrogen assimilation in rapeseed seedlings, and it is essential to apply a suitable glufosinate dose based on the variety grown to minimize adverse effects on crops and environment.


RESUMO: A aplicação de herbicidas é um método eficaz de controle de ervas daninhas para mitigar a perda de produtividade das culturas. No entanto, o uso excessivo de herbicidas pode causar toxicidade em plantas não alvo. O presente estudo avaliou os efeitos da dose recomendada para aplicação agrícola (0.45 kg ha-1) e dose excessiva (0.90 kg ha-1) de glufosinato no desempenho fotossintético e assimilação de nitrogênio das variedades de colza D148 e Zhongshuang 11 (ZS11). Ambas as concentrações de glutamato diminuíram significativamente o teor de clorofila e compostos azotados, exceto a prolina livre, e a atividade de síntese da glutamina e de síntese de glutamato, e aumentaram a atividade de desidrogenase do ácido glutâmico em ambas as variedades. Quando a concentração de glifosato foi 0.45 kg ha-1, a assimilação de azoto das duas variedades diminuiu, o que indicou que a dosagem recomendada de glifosato inibiu a assimilação de azoto das duas variedades de colza. Entretanto, a taxa fotosintética líquida do D148 aumentou enquanto o do ZS11 diminuiu, o que significa que o D148 é mais tolerante a dose recomendada de glifosato do que o ZS11. A dose de 0.90 kg ha-1 de glifosato foi prejudicial para as mudas de duas variedades de colza. Em geral, os nossos resultados indicam que o uso excessivo de glufosinato inibe a taxa fotossintética e a assimilação de nitrogênio em mudas de colza, sendo essencial aplicar uma dose adequada deste herbicida com base na variedade cultivada para minimizar os efeitos adversos nas culturas e no meio ambiente.

ABSTRACT

Objective:To inhibit the Itch gene expression of T-lymphocytes and investigate the immune activity of T lymphocytes using ultrasound-targeted microbubble destruction (UTMD).Methods:T lymphocytes were separated by magnetic bead, and to establish an Itch gene targeted short hairpin RNA (shRNA) plasmid. There were three groups in this study: ①experimental group, Itch-shRNA plasmid-SonoVue microbubbles combined with ultrasound irradiation; ②control group, negative control shRNA plasmid-SonoVue microbubbles combined with ultrasound irradiation; ③blank group, untreated. Forty-eight hours after UTMD transfection, transfection efficiency was detected and analyzed by fluorescence microscopy and flow cytometry, the expression of Itch protein was measured with Western blotting.Seventy-two hours after UTMD transfection, the secretion of interleukin 2 (IL-2) and interferon γ (IFN-γ) in the cell supernatant were measured by enzyme-linked immunosorbent assay (ELISA). One-way ANOVA was used to compare the differences between groups, and LSD- t test was used to compare the differences between groups. Results:The UTMD mediated shRNA transfection rate was 52.3%, and the relative expression levels of Itch protein in the experimental group, control group and blank group were 0.301±0.080, 0.773±0.101 and 0.719±0.090, respectively, and the difference was statistically significant( F=24.441, P<0.01). The Itch gene expression can be effectively suppressed in the experimental group. Seventy-two hours after transfection, the concentrations of IL-2 in the experimental group, control group and blank group were (417.3±37.1)ng/L, (158.7±17.3)ng/L and (147.0±10.2)ng/L, respectively, and the difference was statistically significant( F=118.701, P<0.001) and the concentrations of IFN-γ were (168.3±12.1)ng/L, (74.3±3.7)ng/L and (74.6±7.1)ng/L, respectively, and the difference was statistically significant ( F=126.833, P<0.001). The expression levels of IL-2 and IFN-γ in the experimental group were significantly higher than those in the control group and the blank group. Conclusions:UTMD mediated shRNA transfection can significantly decrease the expression of Itch and promote immune activity of T lymphocyte.

ABSTRACT

Objective:To evaluate the clinical effect of self-made Wenfei-Quyu-Huatan Decoction combined with conventional western medicine in the treatment of acute attack of chronic obstructive pulmonary disease (COPD). Methods:A total of 103 patients with acute attack of COPD who were admitted to our hospital from January 2018 to April 2019 were divided into control group (51 cases) and the intervention group (52 cases) according to the random number table method. The control group was treated with conventional western medicine, and the intervention group was treated with self-made Wenfei-Quyu-Huatan Decoction based on control group. Both groups were treated for 14 days. TCM syndrome scores were recorded before and after the treatment, the FVC, FEV1 and FEV1/FVC were detected by pulmonary function meter, the content of TNF-α, IL-8 and CRP in serum were detected by ELISA, and adverse reactions during the treatment were recorded and clinical efficacy was evaluated. Results:The total effective rate was 88.5% (46/52) in the intervention group and 70.6% (36/51) in the control group, and the difference between the two groups was statistically significant ( χ2=5.067, P=0.024). After the treatment, the TCM syndrome score of the intervention group was significantly lower than the control group (7.3 ± 0.9 vs. 12.9 ± 1.2; t=27.979, P<0.01); FVC (2.69 ± 0.35 L vs. 2.12 ± 0.24 L, t=9.621), FEV1 (1.58 ± 0.22 L vs. 1.13 ± 0.18 L, t=11.349) and FEV1/FVC (0.59 ± 0.07 vs. 0.53 ± 0.08, t=4.053) in the intervention group were significantly higher than the control group ( P<0.01); the serum IL-8 (51.48 ± 6.27 ng/L vs. 69.27 ± 7.32 ng/L, t=13.255), CRP (2.12 ± 0.46 mg/L vs. 3.51 ± 0.53 mg/L, t=14.223) and TNF-α (41.38 ± 6.22 ng/L vs. 52.24 ± 7.29 ng/L, t=8.139) level were significantly lower than the control group ( P<0.01). During the treatment, the incidence of adverse reactions was 11.8% (6/51) in the control group and 15.4% (8/52) in the intervention group, and there was no significant difference between the two groups ( χ2=0.321, P=0.571). Conclusions:The self-made Wenfei-Quyu-Huatan Decoction combined with the conventional western medicine can improve the clinical symptoms, lung function and reduce the level of inflammatory cytokines in patients with COPD acute attack.

ABSTRACT

With the development of liquid biopsy technology, plasma cell-free DNA (cfDNA) becomes one of the research hotspots. Whole-genome bisulfite sequencing of plasma cell-free DNA has shown great potential medical applications such as cancer detection. However, the practical stability evaluation is still lacking. In this study, plasma cell-free DNA samples from two volunteers at different time were collected and prepared for sequencing in multiple laboratories. The library preparation strategies include pre-bisulfite, post-bisulfite and regular whole-genome sequencing. We established a set of quality control references for plasma cell-free DNA sequencing data and evaluated practical stability of blood collection, DNA extraction, and library preparation and sequencing depth. This work provided a technical practice guide for the application of plasma cfDNA methylation sequencing for clinical applications.

PURPOSE@#To compare the efficacy and safety of recombinant human bone morphogenetic protein (rhBMP) and iliac crest autograft in the fusion treatment of lumbar spondylolisthesis.@*METHODS@#The studies using randomized controlled trials to compare the rhBMP with iliac crest autograft in the treatment of lumbar spondylolisthesis were retrieved from Embase, Pubmed, ProQuest dissertations & theses (PQDT), China national knowledge infrastructure (CNKI), Chinese Biomedical Database, Wanfang Data, Cochrane Library (from March 1998 to March 2018). Postoperative fusion rate, clinical success rate, postoperative intervertebral height, complications, operation time, blood loss and duration of hospitalization were chosen as the outcome indicators. Methodological quality of the trials was critically assessed, and relevant data were extracted. Statistical software Revman 5.3 was used for data-analysis.@*RESULTS@#Eleven articles were included in the meta-analysis. The results showed that, comparing the efficacy of rhBMP with iliac crest autograft, statistical significance was found in the 24-month fusion rate post operation [95% CI (1.38, 24.70), p = 0.02] and operation time [95% CI (-14.22, -2.08), p = 0.008]. There is not sufficient evidence for statistical differences in the remaining indicators.@*CONCLUSION@#The current literature shows rhBMP is a safe and effective grafting material in the treatment of lumbar spondylolisthesis. Further evidence is dependent on the emergence of more randomized controlled trials with higher quality and larger sample sizes in the future.

ABSTRACT

Objective To investigate a new clinical classification of hepatolithiasis based on biliary anatomy and pathology in order to guide clinical decision-making.Methods The cases of hepatolithiasis admitted to our hospital from Mar 2015 to Aug 2018 were classified into six types and an additional type according to biliary anatomy,stone site and extent,liver pathological changes,and Oddi sphincter function.Results Of the214 cases of hepatolithiasis,5 cases (2.3%) were type Ⅰ;41 cases (19.2%) were type Ⅱ;76 cases (35.5%) were type Ⅲ;17 cases (7.9%) were type Ⅳ;29 cases (13.5%) were type Ⅴ;46 cases (21.5%) of type Ⅵ.The most common complication was incision infection (28 cases,13.1%),liver failure 0,and perioperative death 0.During the follow-up from 3 months to 3 years,there were 14cases of stone recurrence,the recurrence rate was 7.6%.Conclusion This classification scheme and the "individualized precise anatomical combined hepatectomy or segmental hepatectomy" based on this classification is safe and effective while enhancing the cure rate.

ABSTRACT

Objective To compare the efficacy and safety between minimally invasive plate osteosynthesis (MIPO) and open reduction and internal fixation (ORIF) in the treatment of humeral shaft fracture.Methods Case-control studies and randomized clinical trials comparing MIPO with ORIF in the treatment of humeral shaft fracture from January 2010 to June 2018 were retrieved from PubMed Library,Cochrane Library,Embase Library,China National Knowledge Internet and Wanfang Data.Methodological quality of the studies and trials was critically assessed.REVMANS.3 was used for data analysis.The 2 groups of patients were compared in terms of University of California,Los Angeles shoulder rating scale (UCLA),Mayo Elbow Performance Score (MEPS),union time,nonunion rate and complications.Results A total of 452 patients from 9 articles were included.There were 216 cases in the MIPO group and 236 ones in the ORIF group.The Meta analyses showed that the MIPO group had a significantly higher UCLA score (WMD =0.36,P=0.03),significantly lower incidences of complications (OR =-0.15,P < 0.05) and iatrogenie radial nerve palsy (OR =0.24,P < 0.05),and significantly shorter union time (SMD =-0.36,P =0.02) than the ORIF group.There were no significant differences between the 2 groups in MEPS (WMD =-0.48,P =0.43) or nonunion rate (OR =0.45,P =0.11).Conclusion MIPO may be a better choice for humeral shaft fracture than ORIF in regards to postoperative shoulder functions,union time,and incidences of complications and iatrogenic radial nerve palsy.

ABSTRACT

Present research aims to investigate the repairing effect of polylactic acid-trimethylene carbonate/GNDF slowrelease catheter on the injured femoral nerve fiber. Adult SD male rats as the subjects were divided into two groups, the GDNF group and the control group, and received the surgery to remove the nerve from the exposed left femoral nerves. Thereafter, rats in the GNDF group and the control group received the GNDF or normal saline, and we evaluated the changes in rats, including the morphological, functional and electrophysiological changes of regenerated nerves. Regenerated axons were found in each group, but enormous regeneration of axons was only identified in GDNF group. Further analysis showed that: At the 4th, 8th and 12th weeks, areas of the regenerated nerves in GDNF group were [0.95 +/- 0.06] mm2, [1.14 +/- 0.07] mm2 and [1.22 +/- 0.06] mm2, respectively; in the control group, these were [0.15 +/- 0.01] mm2, [0.25 +/- 0.07] mm2 and [0.52 +/- 0.05] mm2, respectively. These showed that the outcome of GDNF group was superior to that of control group. In GDNF group, quantities of the myelinated fiber were [0.8119×104 +/- 0.0637×104], [1.3371×104 +/- 0.0460×104] and [1.7669×104 +/- 0.0542×104]; while in control group, these were [0.2179×104 +/- 0.0097×104], [0.3490×104 +/- 0.0329×104] and [0.7737×l04 +/- 0.0788×104]. Again, these results also indicated that the outcome of GDNF group was superior to that of the control group [p<0.05]. In GDNF group, the average diameters of myelinated fibers were [2.25 +/- 0.17] micro m, [2.42 +/- 0.14] micro m and [2.80 +/- 0.10] micro m, which were significantly better than [1.24 +/- 0.08] micro m, [1.43 +/- 0.14] micro m and [1.82 +/- 0.14] micro m in the control group. Degrees of fiber myelination in the GDNF group were [0.71 +/- 0.03], [0.64 +/- 0.03] and [0.6l +/- 0.0l], respectively, which were also significantly higher than [0.02 +/- 0.01], [0.04 +/- 0.01] and [0.06 +/- 0.02] in the control group [p<0.01]. At the 12th week after surgery, HE staining was performed to observe the histological changes in quadriceps femoris for evaluation of atrophy in each group. In the GDNF group, significant amelioration was found in the atrophy of quadriceps femoris with an average area of myofiber of [84.95 +/- 3.92] %, while the area of the control group was [57.95 +/- 5.78] %, suggesting that the outcome of the GDNF group was better than that of the control group [p<0.05]. Electrophysiological examination of nerves was employed to detect the recovery of neurological functions after repair of nerve defect. At the 4th, 8th and 12th weeks after surgery, CMAP amplitudes in the GDNF group were [9.34 +/- 0.52] mV, [14.40 +/- 0.69] mV and [19.18 +/- 0.48] mV, significantly better than [0.39 +/- 0.07] mV, [1.44 +/- 0.41] mV and [9.27 +/- 0.40] in the control group [p<0.01]. Polylactic acid-trimethylene carbonate/GNDF slowrelease catheter can accelerate the functional recovery of injured nerves, thus promoting the regeneration efficiency of femoral nerves

ABSTRACT

OBJECTIVE@#This study aims to investigate the effect of neurotrophin 3 (NT-3) on the osteogenic differentiation of human dental follicle cells (hDFCs).@*METHODS@#hDFCs were isolated and cultured in vitro. Immunocytochemical staining was used to identify the origin of hDFCs. The effects of different NT-3 concentrations on hDFCs proliferation were detected by using CCK-8 assay. The alkaline phosphatase (ALP) activities and mRNA expression levels of bone morphogenetic protein-2 (BMP-2) and osteocalcin (OCN) were determined to investigate the effects of NT-3 on hDFCs osteogenesis. The difference in the number of mineralized nodules was detected using alizarin red staining.@*RESULTS@#Vimentin and cytokeratin staining results showed that hDFCs originated from the mesenchymal cells. NT-3 exerted no evident effect on hDFCs proliferation. The ALP activity and the BMP-2 and OCN mRNA expression levels of hDFCs were significantly improved under treatment with different NT-3 concentrations (25, 50, and 100 ng·mL ⁻¹) compared with those in the control group. BMP-2 and OCN mRNA relative expression levels of hDFCs reached the highest when the NT-3 concentration was 100 ng·mL ⁻¹. The number of mineralized nodules reached the maximum when the hDFCs were treated with 50 and 100 ng·mL ⁻¹ NT-3.@*CONCLUSIONS@#Appropriate mass concentration of NT-3 can promote the osteogenic differentiation of hDFCs.

ABSTRACT

Objective To study the association between the expression of periostin and the occurrence and progression of knee osteoarthritis (OA) in synovial fluid.Methods The expression level of periostin in the synovial fluid of healthy people and patients with different stages of OA was tested.Furthermore,60 surgical-induced OA rat model were divided into two groups,the sham operation group had only implemented slit suture,and the OA model group had one side anterior cruciate ligament transected.The expression of periostin in intra-articular injection samples were analyzed at 1,2,4,8,12 week.Fluorescence molecular tomography (FMT) were performed after surgery at 4,8,12 week on the surgery knee.Gross morphologic lesions on the tibial plateau in rats were visualized by India ink staining,toluidine blue staining,cartilage permeation test.The synovium were visualized by HE staining and periostin were detected by immunohistoc hemistry.The measurement data were compared by one factor analysis of variance test.Results The expression of periostin in cartilage was lower in late-stage OA than the one from normal and early-stage OA (F=13.95,P<0.01).The FMT showed that there was no obvious change in the extent of chronic inflammation in the sham operation group,and the chronic inflammatory degree of the OA model group gradually increased as time went on.Toluidine blue staining and cartilage permeation test showed that the cartilage degeneration in rat model of OA became more and more serious with time.There was no stastically significant difference of the periostin in control groutp at different time stage (F=0.67,P=0.53).The periostin in the intra-articular increased at first and then decreased with the development of OA (F=11.0,P<0.05).HE staining of synovial tissue showed that the degree of synovial hyperplasia was consistent with the degree of degeneration of joints.With the extension of time,the expression of periostin in synovial tissue increased gradually.Conclusion The expression of periostin in human synovial fluid is low in normal knee joint,increases in early and middle stages,and decreases in late stage.The rat model indicates that the expression of periostin increases first and then decreases with the development of OA,but the expression in synovium increases gradually with the development of OA.The increased expression of periostin in synovial fluid may serve as an early diagnostic marker for OA and downregulation of the periostin may be a start marker for the late OA.

ABSTRACT

Objective To investigate the effect of POSTN protein on the proliferation of chondrocytes of tibial plateau in old rats.Methods Cartilage cells collected from the tibial plateau of old rats were cultured in vitro to the third generation.Then the cells were divided into 3 groups:POSTN group,PBS group and POSTN antibody group.The proliferations of the three groups at 24 h,48 h and 72 h were determined by EDU method.The expression of Notch1 protein was detected by immunohistochemistry in all groups at the same time.Female 20-month-old Sprague-Dawley rats were randomly divided into 3 groups:POSTN protein injection group,Phosphate Buffered Saline (PBS) injection group and POSTN antibody injection group.Twelve weeks after the operation,related reagents were injected 3 times consecutively at day 1,day 3,day 5 and EDU was injected into joints at day 1.At 2 weeks after injection,the rats were killed and the knee tibial plateau was taken to observe the proliferation of the cartilage cells.Results At 24 h,there were differences between the three groups O(F=27.32,P=0.017).The proliferation rates of POSTN group [(23±8)%] and PBS group [(21±10)%] were higher than that of POSTN antibody group (16±5)(P=0.003,P=0.011).At 48 h,there were differences between the three groups (F=35.34,P<0.01).The proliferation rate of POSTN group [(36±11)%] was higher than that of the other groups [(22±6)%],(18±6)%(P=0.021,P<0.01).At 72h,there were differences between the three groups (F=52.62,P=0.000).The proliferation rate of POSTN group [(56±17)%] was the highest one,the proliferation rate of PBS group [(31±8)%] was the medium,and the POSTN antibody group [(26±7)%] was the lowest one (all P<0.05).As for Notch1 protein expression in chondrocytes,there were differences between the three groups (F=26.72,P<0.01).The Notch1 protein was the most frequently expressed in POSTN protein-injection group and the least in the anti-POSTN group.In rats,the proliferation rate of the chondrocytes in the medial tibia plateau of the knee of POSTN protein injection group [(36±14)%],which was the highest,and that of the POSTN antibody injection group [(10 ±4)%] was the lowest (all P<0.05).Conclusion POSTN protein can promote the proliferation of chondrocytes knee OA rats.POSTN antibody injection has been shown to induce the proliferation of chondrocytes.The POSTN protein may promote the proliferation of chondrocytes by activating the Notch signaling pathway.
